Glutathionperoxidase 1

Schädliche Formen des Sauerstoffs werden als reaktive Sauerstoffspezies (ROS) bezeichnet. ROS entstehen natürlicherweise in allen Zellen als Nebenprodukte verschiedener Stoffwechsel­vorgänge (z. B. Zellatmung) oder als Entzündungsreaktion durch Immunzellen. Gase wie Ozon, UV-Strahlung oder Luftpartikel können ebenfalls zur Bildung von ROS führen. ROS führen zu oxidativem Stress, können alle Strukturen der Zelle schädigen und zum Zelltod führen.

 

Eine wichtige Rolle bei der Entgiftung von ROS spielen die Glutathionperoxidasen (GPX). Sie übertragen Wasserstoff von Glutathion auf organische Hydroperoxide (R–O–OH) oder Wasserstoffperoxid (H2O2) und schützen die Zellen so gegen oxidative Schäden. Es gibt verschiedene GPX-Isoformen, die sich hinsichtlich ihrer zellulären Lokalisation und ihrer Substratspezifität unterscheiden.

 

Die zelluläre GPX1 ist ubiquitär vorhanden und besteht aus vier identischen Monomeren mit je einem Selen im aktiven Zentrum. Bei Selenmangel wird weniger GPX1-Enzym gebildet. Eine Selensubstitution erhöht die GPX1-Aktivität. Die GPX1-Aktivität unterliegt interindividuellen Schwankungen, die u. a. vom Geschlecht, Alter, Nahrungsergänzungs- und Raucherstatus abhängen(Bolzán, Bianchi and Bianchi, 1997).

 

Außerdem führt ein genetischer Polymorphismus im GPX1-Gen zu einer um ca. 10 % verminderten Enzymaktivität (Bastaki et al., 2006; Ravn-Haren et al., 2006; Takata et al., 2012). Diese ist Folge eines Basenaustauschs von Cytosin zu Thymin an der Position c.599C>T (rs1050450) im GPX1-Gen mit dem Aminosäureaustausch von Prolin zu Leucin (Pro200Leu). In der europäischen Bevölkerung tragen etwa 47,6 % an der Position c.599 homozygot Cytosin (CC). 43,8 % der Bevölkerung haben mit CT und 8,5 % mit TT einen verminderten oxidativen Schutz (Vogel et al., 2004). Personen mit einer genetisch bedingt geringeren GPX1-Aktivität erkranken häufiger an diabetischen Polyneuropathien, neurodegenerativen Krankheiten, Brust-, Lungen-, Dickdarm- und Prostatakrebs sowie Erkrankungen, die mit oxidativem Stress assoziiert sind (z. B. kardiovaskulärer Erkrankungen). Eine Hemmung der GPX1 Aktivität wird bei erhöhten Selenplasmakonzentationen nur für Träger der homozygot unveränderten Genvariante (CC) beschrieben (Miller et al., 2012).
